ABSTRACT

Objective:To explore the prognostic predictive value of detecting minimal residual disease (MRD) after 2 courses of hypomethylating agents (HMA) combined with low-dose induction chemotherapy in patients with acute myeloid leukemia (AML).Methods:The data of 43 newly diagnosed AML patients treated by HMA combined with low-dose induction chemotherapy in Jingjiang People's Hospital of Jiangsu Province from January 2016 to January 2021 were retrospectively analyzed, and the bone marrow MRD levels were detected by multiparametric 10-color flow cytometry (MFC) after 1 course and 2 courses of chemotherapy. Patients were divided into three groups according to MRD levels: the group with negative MRD after 1 course of induction chemotherapy (MRD-1 group), the group with negative MRD after 2 courses of induction chemotherapy (MRD-2 group), and the group without negative MRD after 2 courses of induction chemotherapy (MRD+ group). Kaplan-Meier method was used to draw the progression-free survival (PFS) and overall survival (OS) curves of all patients and each group, and log-rank test was performed to compare them; the influencing factors for OS were analyzed using univariate and multivariate Cox proportional hazards models.Results:Among the 43 patients, 17 patients (39.5%) were in the MRD-1 group, 14 patients (32.6%) were in the MRD-2 group, and 12 patients (27.9%) were in the MRD+ group. There were no statistical differences among the 3 groups in gender, age, hemoglobin level at initial diagnosis, white blood cell count, platelet count, lactate dehydrogenase level, disease subtype, WT1 expression, karyotype, and genetic risk stratification (all P > 0.05). The median follow-up was 15 months (1-67 months). Survival analysis showed a median OS time of 21 months (95% CI 15 months -not reached) in 43 patients and a median PFS time of 12 months (95% CI 9-18 months) in 29 patients included in the PFS analysis; PFS and OS in the MRD-1 and MRD-2 groups were better than those in the MRD+ group (all P < 0.01), and the differences in PFS and OS between the MRD-1 and MRD-2 groups were not statistically significant (both P > 0.05); the median PFS time was 5 months (95% CI 2 months-not reached) in the MRD+ group, the median PFS time was 15 months (95% CI 7 months-not reached) in the MRD-1 group, and the median PFS time was 18 months (95% CI 11 months-not reached) in the MRD-2 group; the median OS time in the MRD+ group was 9 months (95% CI 7 months-not reached), the median OS time was not reached in the MRD-1 group, and the median OS time was 38 months (95% CI 38 months-not reached) in the MRD-2 group. Multivariate Cox regression analysis showed that age ( HR = 1.080, 95% CI 1.004-1.160, P = 0.038), MRD status (MRD-1 vs. MRD+: HR = 0.125, 95% CI 0.031-0.507, P = 0.004; MRD-2 vs. MRD+: HR = 0.146, 95% CI 0.037-0.577, P = 0.006) were independent influencing factors for OS in AML patients. Conclusions:The survival is good in AML patients with MRD negative conversion after both 1 course and 2 courses of HMA combined with low-dose induction chemotherapy, and both are better than that in patients with positive MRD after 2 courses of chemotherapy.

ABSTRACT

Objective:To investigate the correlation of excessive platelet (Plt) recovery at the first time of achieving morphologic complete remission (CR) after induction chemotherapy with minimal residual disease (MRD) and the clinical features of acute myeloid leukemia (AML).Methods:The clinical data of newly-treated 57 AML patients (except for acute promyelocytic leukemia) who achieved CR after induction chemotherapy in Jinjiang People's Hospital from January 2016 to December 2021 were retrospectively analyzed. A total of 57 newly diagnosed adult AML patients were divided into excessive Plt recovery group (Plt recovery>350×10 9/L) and normal Plt recovery group [Plt recovery: (100-350)×10 9/L] according to the Plt recovery. Meanwhile, the MRD was analyzed by using multiparameter flow cytometry (MFC) in patients achieving CR after receiving 1 course of standard treatment regimen or 1-2 courses of demethylation drugs combined with pre-conditioning regimen. The clinical features and negative rate of MRD between the two groups were compared. Results:Among 57 CR patients, 31 (54.4%) patients had CR with excessive Plt recovery and MFC-MRD negative rate was 67.7% (21/31); 26 (45.6%) had CR with normal Plt recovery and MFC-MRD negative rate was 38.5% (10/26); and the difference in the proportion of MRD negative patients between the both groups was statistically significant ( χ2 = 4.89, P = 0.027). There were no statistically differences in the proportions of patients with different gender, age, WBC at initial diagnosis, Plt, chemotherapy regimen and risk degree classification between the two groups (all P > 0.05). Conclusions:In AML patients, excessive Plt recovery at the first time of achieving morphologic CR after induction chemotherapy is associated with negative MRD, which has a certain value in the judgement of therapeutic effect.

ABSTRACT

Objective:To explore the clinical efficacy and treatment costs of different regimens for newly diagnosed acute promyelocytic leukemia (APL).Methods:The clinical data of 37 newly diagnosed APL patients admitted to Jingjiang People's Hospital from January 2011 to December 2020 were retrospectively analyzed. They received different induction therapy regimens and consolidation therapy after achieving complete remission (CR). Among them, 11 patients received induction chemo therapy with all-trans retinoic acid (ATRA) combined with anthracycline, which was consolidated with ATRA combined with chemotherapy after CR (ATRA+chemotherapy group); 13 patients were treated with ATRA combined with arsenite acid (ATO) and anthracycline, which was consolidated with ATRA combined with chemotherapy after CR(ATRA+ATO+ chemotherapy group). The other 13 patients received double induction therapy of ATRA combined with ATO, which was consolidated with ATRA combined with ATO after CR (ATRA+ATO double induction group). The clinical efficacy and treatment costs of newly diagnosed APL patients in 3 groups were analyzed.Results:There were 10, 12, 12 patients with newly diagnosed APL achieving CR, respectively in ATRA+chemotherapy group, ATRA+ATO+chemotherapy group, ATRA+ATO double induction group, and the difference was statistically significant ( P > 0.05). The differences of hematological, cardiac, gastrointestinal adverse reactions and infection incidence in the 3 groups were not statistically significant (all P > 0.05). The costs of induction therapy in ATRA+chemotherapy group, ATRA+ATO+chemotherapy group and ATRA+ATO double induction group were (73 755±4 820) yuan, (74 101±5 097) yuan, (52 944±4 099) yuan, respectively; the costs of consolidation treatment were (26 366±2 497) yuan, (25 801±2 528) yuan, (19 674±1 940) yuan, and the treatment time was (41±4) d, (39±4) d, (34±3) d, respectively; and the differences were statistically significant ( F value was 84.77, 31.90, 9.62, all P < 0.001). Conclusion:The chemotherapy-free therapy regimen of induction and consolidation with ATRA and ATO has the advantage of high cost-effectiveness with no significant difference in clinical efficacy.

ABSTRACT

OBJECTIVE To identify an d analyze chemical cons tituents of Pleione yunnanensis with origin of Pleione yunnanensis. METHODS UPLC-Q-Exactive-Plus-Orbitrap-MS was adopted. The determination was performed on Hyperdil GOLD column with mobile phase consisted of 0.1% formic acid solution-0.1% formic acid acetonitrile solution (gradient elution )at the flow rate of 0.3 mL/min. The column temperature was set at 40 ℃,and sample size was 2 µL. The electrospray ion source was adopted,and the scanning range was m/z 100-1 500,and the scanning mode was positive and negative ion exchange mode of full scan+ddMS2. The structure of chemical constituents were determined by using Compound Discoverer 3.1 software,comparing with mzCloud,PubChem network database and OTCML ,on the basis of reference substance and published literatures. RESULTS & CONCLUSIONS A total of 42 chemical constituents were identified (positive ion mode has 24,negative ion mode has 27), including 13 benzyl succinate glycosides (such as dactylorhin C ,coelovirin A ,militarine),4 phenol glycosides (such as adenosine , guanosine,gastrodin),3 alkaloids(choline,betaine,berberine),and one flavonoid (nobiletin),7 aromatics(such as DL-lysine , DL-arginine,DL-glutamine),one sugar (sucrose),3 benzenes(shancigusin H ,shancigusin H isomer ,batatasin Ⅲ)and 10 others (such as p-methoxybenzoic acid ,monomethyl dodecanedioate ,diphenylamine). Glucose oxybenzyl and some small mole cules are easy to be lost in the cleavage of benzyl succinate glycosides;glycosyl is easy to be lost in the cleavage of phenol glycosides;the cleavage of alkaloids mainly manifest as the cleavage and loss of small molecular substituents ;demethyla- tion reaction is occurred in most flavonoids.

ABSTRACT

OBJECTIVE To establis h the fingerprint of Cynanchum auriculatum,to conduct its chemical pattern recognition analysis,and to determine the contents of four components at the same time. METHODS High performance liquid chromatography (HPLC)method was adopted. The determination was performed on ACE UExcel C 18 column with mobile phase consisted of acetonitrile-0.1% phosphoric acid solution (gradient elution )at the flow rate of 1.2 mL/min. The determination wavelength was set at 210 nm,and the column temperature was 40 ℃. The sample size wa s 10 μL. Taking qingyang shengenin as the reference ,HPLC fingerprints of 16 batches of C. auriculatum medicinal materials were drawn and similarity was evaluated by using the Similarity Evaluation of Chromatographic Fingerprints of Traditional Chinese Medicine (2012 edition), and the common peaks were determined. SPSS 26.0 software andSIMCA 14.0 software were used for cluster analysis ,principal component analysis and orthogonal partial least squares- discriminant analysis. The differential components affecting the quality of C. auriculatum were screened by taking the value of variable importance in projection (VIP)greater than 1 as the standard ;same HPLC method was used to determine the contents of syringic acid ,acyl asclepiadelenin ,baishouwubenzophenone and qingyang shengenin. RESULTS There were 29 common peaks in 16 batches of C. auriculatum ,with a similarity of 0.723-0.998. Four common peaks were identified ,namely syringic acid (peak 7),acyl asclepiadoidin (peak 9),baishouwubenzophenone(peak 13)and qingyang shengenin(peak 15). The results of cluster analysis showed that 16 batches of C. auriculatum could be clustered into three categories ,among which S 1 were grouped into one category,S3 were grouped into one c ategory,S2,and S 4-S16 were grouped into one category. The results of principal component analysis showed that the cumulative variance contribution rate of the five principal components was 88.706%,and the classification results were consistent with the results of cluster analysis. The results of orthogonal partial least squares-discriminant analysis showed that the common peaks (from large to small )with VIP value greater than 1 were peak 20,peak 10,peak 25,peak 12, peak 15(qingyangshengenin),peak 21,peak 14,peak 16,peak 26,peak 22 and peak 17. The linear ranges of syringic acid,acyl asclepterin,baishouwubenzophenone and qingyangshengenin were 0.715 3-45.778 0,2.379 4-152.281 0,0.642 0- 41.085 0,14.541 6- 930.662 0 µg/mL respectively (all R 2>0.999). The quantitative limits were 0.357 7,0.475 9,0.642 0 and 2.423 6 μg/mL;the detective limits were 0.146 0,0.164 1,0.248 8 and 0.833 3 μg/mL,respectively. RSDs of precision ,repeatability and stability (24 h)tests were less than 3%;the average recoveries were 99.11%(RSD=2.00%,n=9),98.54%(RSD=2.21%,n=9), 96.33%(RSD=2.54%,n=9)and 95.96%(RSD=2.93%,n=9);the contents were 17.12-147.80,95.23-583.10(S8 below the quantitative limit ),16.91-210.88 and 211.68-3 587.15(S1 below the quantitative limit )μg/g,respectively. CONCLUSIONS Established HPLC fingerprint and the method of content determination are stable ,reliable,accurate and reproducible. Combined with analysis of chemical pattern recognition ,it can be used for the quality control of C. auriculatum .

OBJECTIVE@#To explore the genetic basis for an individual with a para-Bombay phenotype.@*METHODS@#A proband with mismatched forward and reverse serotypes for the ABO blood group was identified. Weakly expressed ABH blood type antigen on the surface of red blood cells was verified by absorption and release test, and the blood group substances in saliva was detected by sialic acid test. Exons 6 and 7 of the ABO gene and exons of the FUT1 and FUT2 genes were subjected to direct sequencing.@*RESULTS@#The proband was found to be of O type by forward ABO serotyping and AB type by reverse ABO serotyping, though H and substance A and B were detected in her saliva. DNA sequencing revealed that she has harbored c.35C/T, c.328G/A, and c.504delC compound heterozygous variants of the FUT1 gene. Haploid analysis showed that her FUT1 genotype was h328A/h35T+504delC, which has been uploaded to the NCBI website (No. MW323551).@*CONCLUSION@#The para-Bombay phenotype of the proband may be attributed to the novel compound heterozygous variants including c.504delC of the FUT1 gene, which may affect its function by altering the activity of FUT1 glycotransferase.

ABSTRACT

OBJECTIVE To improve th e quality standard for Gerbera piloselloides. METHODS The properties of G. piloselloides were observed and microscopic identification was conducted for powder. The moisture ,total ash ,acid-insoluble ash and ethanol-soluble extract were detected according to the method stated in 2020 edition of Chinese Pharmacopoeia (part Ⅳ). The contents of nodakenin,luteolin-7-O-β-D-lutinoside,luteoloside,apigenin-7-O-β-D-lutinoside,apigenin-7-O-β-D-glucopyranoside and marmesin were determined by high performance liquid chromatography method. RESULTS G. piloselloides were shrunken ,densely covered with thick white cotton wool ,with many fibrous roots ;the surface was taupe or gray-brown ;its texture was brittle and easy to break ;the cross section was yellow-white ,and there was an obvious small wooden heart in the center. Its powder was tan , and non-glandular hairs ,stone cells ,calcium oxalate cubes ,ducts,fibers could be seen unde r microscope. The measured values of moisture,total ash ,acid-insoluble ash and alcohol-soluble extract , for 15 batches of samples were 8.63%-11.34%,10.39%-14.93%, 3.29%-6.37% and 9.03%-15.02%,respectively;average values were 10.01%,12.26%,4.61%,12.36%. The linear ranges of nodakenin,luteolin-7-O-β-D-rutinoside,luteoloside,apigenin- 7-O-β-D-lutinoside,apigenin-7-O-β-D-glucopyranoside and marmesin we re 3.87-154.88,1.64-65.41,1.60-64.00,1.92-76.96, 1.27-50.93,0.40-15.89 μg/mL,respectively(r≥0.999 1). RSDs of precision ,stability(24 h)and repeatability tests were all less than 3%. The average recoveries were 101.88%,100.89%,102.64%,95.75%,96.71% and 103.48%,respectively;RSDs were 0.55%,0.43%,0.34%,0.49%,0.47% and 0.37%,respectively(n=6);the contents of above 6 components were 0.152 7-0.852 2, 0.084 5-0.669 7,0.136 7-0.961 0,0.126 0-1.193 2,0.128 8-1.102 2,0.046 9-0.678 0 mg/g. CONCLUSIONS The established method can be used for the quality control of G. piloselloides . It is preliminarily proposed that the moisture in G. piloselloides is not more than 12.0%;the total ash is not more than 15.0%,the acid-insoluble ash is not more than 6.0%,the alcohol-soluble extract is not less than 9.0%;the contents of luteoloside and apigenin- 7-O-β-D-glucopyranoside are not less than 0.016%.

ABSTRACT

Objective:To investigate the effect of Caprini risk score-based care on the prevention of venous thromboembolism of patients with major orthopedic surgery.Methods:Totally 124 patients with major orthopedic surgery were randomly divided into experimental group and control group, each group contained 62 cases. The control group received routine thrombosis prevention nursing, patients in the experimental group carried out Caprini risk score-based intervention. The rate of VTE was compared between two groups, the levels of D-Dimer as well as thigh circumference was measured after 24 hours of admission, 3 days after surgery, 5 days after surgery and at discharge day. At discharge day, the inpatients satisfaction with nursing service scale was used to assessment the satisfaction with nursing service of inpatients.Results:The rate of VTE in the experimental group was 1.6% (1/62) and 6.5% (4/62) in the control group, the difference was not statistically significant ( χ2 value was 1.876, P=0.171). Three days after surgery, 5 days after surgery and at discharge day, the D-Dimer were (1.26±0.65) μg/ml, (1.27±0.35) μg/ml, (0.96±0.27) μg/ml in the experimental group, 5 days after surgery and at discharge day, the thigh circumference were (29.81±4.15) cm, (28.86±3.40) cm in the experimental group, significantly lower than those in the control group [(1.64±0.58) μg/ml, (1.51±0.55) μg/ml, (1.25±0.44) μg/ml, (31.84±5.22) cm, (30.83±3.91) cm], the difference was statistically significant ( t value was 2.405-4.485, P<0.05). The health education and guidance, personality traits, service quality, discharge guidance scores and total satisfaction scores were 26.31±3.06, 35.73±3.64, 27.89±3.11, 8.47±1.18 and 111.08±6.33, significantly higher than those in the control group (24.15±2.31, 33.92±2.66, 26.27±3.41, 7.42±1.82 and 106.52±5.85), the difference was statistically significant ( t value was 2.750-4.433, P<0.05). Conclusions:Caprini risk score-based nursing care significantly alleviates the hypercoagulable states and relieves the degree of swelling of the affected of patients with major orthopedic surgery, as well as improves patients' satisfaction with nursing work.

ABSTRACT

OBJECTIVE:To study the improvement effects and mechanism of Polygonum orientale flower extract on hypoxia- reoxygenation injury of H 9c2 cardiomyocytes. METHODS :H9c2 cardiomyocytes were divided into normal control group ,model group and low- ,medium- and high-concentrations groups of P. orientale flower extract (20,40,80 μg/mL). Except for normal control group ,other groups were given 800 μmol/L CoCl2 to induce hypoxia-reoxygenation injury model. Cell apoptosis was observed. The levels of Ca 2+(in cytoplasm ),mitochondrial membrane potential (MMP),ATP enzyme (Na+-K+-ATP enzyme ,Ca2+-Mg2+-ATP enzyme) activities, the ratio of cytochrome c (Cyto c ), protein in cytosol to mitochondria ,phosphorylation levels of reperfusion injury salvage kinase (RISK) signaling pathwayrelated protein [protein kinase B (Akt)and extracellular signal regulated kinase 1/2(ERK1/2)] as well as protein expression of HIF- 1 α were detected respectively. In addition,the cells were divided into normal control group ,model group and P. orientale flower extract group (80 μ g/mL),PI3K inhibitor LY294002+CoCl2 group(15 μmol/L LY294002+80 μmol/L ,LY294002+P. orientale flower extract group (15 μmol/L LY294002+80 μg/mL P. orientale flower extract ),MEK inhibitor PD98059+CoCl2 group(25 μmol/L PD98059+800 μmol/L CoCl2),PD98059+P. orientale flower extract group (25 μmol/L PD98059+80 μg/mL P. orientale flower extract ). After cultured by the same method ,the phosphorylation levels of Akt protein and ERK1/2 protein in the cells were measured to verify the activation of P. orientale flower extract to RISK signaling pathway. RESULTS:Compared with model group ,nuclear pyknosis and the number of apoptotic bodies were reduced in different concentrations groups of P. orientale flower extract. ROS level ,Ca2+ level(except for low-concentration group ),MMP,ratio of Cyto c in cytoplasm to Cyto c in mitochondria ,protein expression of HIF- 1α were decreased significantly(P<0.05 or P<0.01); the activity of ATP enzyme (except for the low-concentration group ),Akt protein and ERK 1/2 protein phosphorylation level were significantly increased (P<0.01). After treated with PI 3K inhibitor LY 294002 and MEK inhibitor PD 98059,Akt protein and ERK 1/2 protein phosphorylation level in cadiomyocyte were decreased significantly (P<0.05 or P<0.01). CONCLUSIONS :P. orientale flower extract can improve hypoxia-reoxygenation injury of H 9c2 cardiomyocytes,the mechanism of which may be associated with inhibiting cardiomyocyte apoptosis ,improving ATPase activity ,protecting mitochondria ,regulating RISK signaling pathway related proteins and HIF- 1α protein expression.

ABSTRACT

Early recognition and treatment for early warning electrocardiogram (ECG) of sudden death are very important to prevent and treat malignant arrhythmia and sudden death. Previous studies have found that R-on-T and T wave alternation, and QT interval prolongation are closely related to malignant arrhythmia or sudden death, which are included in the critical value of ECG.By analyzing the ECG characteristics of 4 patients with sudden death, we found that although the causes of the patients were different, there were transient prolongation of QT interval after premature contraction in 12 lead ECG, followed by malignant arrhythmia or sudden death. Thus, we thought that the transient prolongation of QT interval after premature contraction had a high value for warning malignant arrhythmia or sudden death. This phenomenon should be paid enough attention to reduce the risk of sudden death.

ABSTRACT

Neonates are undergoing the transition period from fetal circulation to adult circulation, and the hemodynamic changes are complex.For premature infants(especially very low birth weight infants and extremely low birth weight infants), the cardiovascular systems are immature.Therefore, they are prone to hypotension at early postnatal stage.Hypotension may lead to multiple organ perfusion insufficience, cerebral lesion, and even death.However, it is not always accompanied by hypoperfusion.Before treatment, clinicians should evaluate the hemodynamics comprehensively.At present, there is no consensus on the definition and intervention threshold about hypotension in premature infants.This review summarizes the definitions and hemodynamic monitoring methods of hypotension in premature infants at early postnatal stage to provide references for diagnosis and treatments.

ABSTRACT

Objective:To investigate the mechanism of lncRNA Sox2OT in patients with Alzheimer's disease (AD) induced by A type of β peptide (Aβ1-42).Methods:Rat pheochromocytoma cells (PC12 cells) were selected and treated by Aβ1-42 to establish PC12 cell model.PC12 cells were set as blank group before induction to verify the successful construction of the cell model.The induced PC12 cells were divided into control group, Sox2OT overexpression (p-Sox2OT) group, p-Sox2OT empty vector (p-NC) group, inhibited Sox2OT expression (si-Sox2OT) group and si-Sox2OT empty vector (si-NC) group.The proliferation activity of thiazole blue (MTT) was detected.Flow cytometry was used to detect the cell cycle and apoptosis rate after transfection.Results:MTT results showed that compared with the blank group (99.67±10.50), the cell proliferation rate of the control group (29.33±5.51) was significantly reduced ( t=10.27, P<0.05). RT-qPCR results showed that compared with the control group (0.52±0.06), the Sox2OT mRNA expression level in the p-Sox2OT group (2.19±0.16) was significantly increased ( t=16.93, P<0.05). The mRNA expression level of Sox2OT in the si-Sox2OT group (0.22±0.02) decreased significantly ( t=15.28, P<0.05). Compared with the p-NC group (0.53±0.12), The mRNA expression level of Sox2OT in the p-Sox2OT group (2.19±0.16) was significantly increased ( t=16.25, P<0.05). Compared with the si-NC group (0.51±0.09), the mRNA expression level of Sox2OT in the si-Sox2OT group (0.22±0.02) was significantly decreased ( t=16.93, P<0.05). The difference between the control group, the p-NC group and the si-NC group was not statistically significant ( P>0.05). In addition, the cell proliferation ability of the p-Sox2OT group (145.00±5.12) was significantly higher than that of the si-Sox2OT group (23.33±4.93), control group (55.00±5.00), si-NC group (57.33±8.51) and p-NC group (56.00±5.57) ( t=29.65, 21.78, 27.55, 21.35, all P<0.05). The difference in cell proliferation rate between Control group, p-NC group and si-NC group was not statistically significant ( P>0.05). Cell cycle detection experiments showed that the number of cells in the G1 phase of the p-Sox2OT group was significantly lower than that of the control group and p-NC ( t=9.80, 8.57; both P<0.05), while the number of cells in the G2 phase of the p-Sox2OT group was significantly higher than that of the control group and the p-NC group ( t=11.02, 10.25; both P<0.05). The number of cells in the G1 phase of the si-Sox2OT group was significantly higher than that of the control group and the si-NC group ( t=8.22, 3.11, both P<0.05), while the number of cells in the G2 phase of the si-Sox2OT group decreased significantly, compared with the control group and the si-NC group ( t=6.32, 5.33; all P<0.05). There was no statistically significant difference in cell cycle between the control group, the p-NC group and the si-NC group (both P>0.05). In the S phase, the difference between the p-Sox2OT group and the control group was statistically significant ( t=1.84, P<0.05). The number of cells in the G2 phase of the p-Sox2OT group (19.00±1.00) was significantly higher than that of the si-Sox2OT group (3.33±1.53), the control group (10.00±1.00), si-NC group (8.55±0.73) and p-NC group (7.67±1.53) ( t=14.85, 11.02, 10.23, 10.74, all P<0.05). The apoptosis rate of p-Sox2OT group ((3.66±0.26)%) was lower than that of si-Sox2OT group ((14.25±0.80)%), control group ((8.46±0.44)%), si-NC group ((8.78±0.44)%) and p-NC group ((8.40± 0.21)%) ( t=21.81, 16.27, 20.32, 21.35, all P<0.05). For the apoptosis rate, there was no statistically significant difference between control group, p-NC group and si-NC group( P>0.05). In addition, the expression levels of p-PI3K and p-Akt in the p-Sox2OT group were significantly higher than those in the p-NC group ( P<0.05). Compared with the si-NC group, the expression of p-PI3K and p-Akt in PC12 cells in the si-Sox2OT group was significantly decreased ( P<0.05). Conclusion:lncRNA Sox2OT can promote the proliferation of PC12 cells induced by Aβ1-42 and inhibit apoptosis by regulating the PI3K/Akt pathway.

ABSTRACT

Objective:Exploring tau related disease pattern (tauRDP) in the brain of Alzheimer′s disease (AD) patients based on 18F-APN-1607 PET scan. Methods:18F-APN-1607 PET images were collected from 17 AD patients (6 males and 11 females, age: (61.7±12.3) years, Mini-Mental State Examination (MMSE) score: 17.6±7.9) and 10 normal controls (NC; 6 males and 4 females, age: (61.2±4.7) years) from Huashan Hospital of Fudan University. The scaled subprofile model (SSM) based on principal component analysis (PCA) technique was used to construct the tauRDP. Then the expression value of tauRDP in each sample was calculated. The differences on tauRDP expression values between AD patients and NC were compared by independent-sample t test. Pearson correlation analysis was used to analyze the correlation between tauRDP expression values and MMSE values in AD patients. Results:The tauRDP area mainly included: precentral gyrus, dorsolateral superior frontal gyrus, middle frontal gyrus, inferior frontal gyrus of opercular part, inferior frontal gyrus of triangular part, supplementary motor area, medial superior frontal gyrus, left median cingulate and paracingulate gyri, right cuneus, superior occipital gyrus, middle occipital gyrus, postcentral gyrus, superior parietal gyrus inferior parietal, but supramarginal and angular gyri, supramarginal gyrus, angular gyrus, precuneus and middle temporal gyrus. There were significant differences ( t=4.395, P<0.001) between AD group (12.6±8.0) and NC group (0.0±1.0) in tauRDP expression values. The tauRDP expression values were correlated with MMSE values in AD group significantly ( r=-0.566, P=0.018). Conclusions:TauRDP established basing on SSM/PCA method can be used to quantitatively express the abnormal spatial distributions of tau deposition. Expression value of tauRDP has the potential to initially assess the severity of AD.

ABSTRACT

OBJECTIVE:To s tudy the intestinal absorption differences of 6 kinds of active constituents of Polygonum orientale (kaempferol,isokaempferol,vitexin,protocatechuic acid ,kaempferol-3-O-β-D-glucoside and quercetin )in normal and myocardial ischemia(MI)model rats. METHODS :UPLC-MS/MS method was adopted to determine the contents of 6 active components in the intestinal circulatory perfusion fluid. Totally male SD 80 rats were divided into normal group and model group ,with 40 rats in each group. Model group was given isoproterenol hydrochloride (50 mg/kg) subcutaneously to induce MI model;normal group was given constant volume of normalsaline, once a day , for consecutive 2 days. 24 h after successful molding ,normal group and model group received in-situ intestinal circulatory perfusion experiment. The effects of different concentration s of P. orientale extract(5.0,10.0, 20.0 mg/mL),different intestinal segments (duodenum,jejunum,ileum,colon),P-glycoprotein(P-gp)inhibitors(verapamil) and bile on the intestinal absorption of each constituent were explored. RESULTS :The linear ranges of concentrations of kaempferol, isokaempferol, vitexin, protocatechuic acid , kaempferol-3-O-β-D-glucoside and quercetin were 3.15-50.40, 3.21-51.31,1.63-52.43,1.60-50.94,1.31-20.97,8.07-129.25 µg/mL(r>0.999). The lower limits of quantification were 7.86, 8.45,6.52,4.00,3.28,16.14 ng/mL,respectively. RSDs of precision ,matrix effect and stability tests were all lower than 11%; the accuracy were 85.64%-107.65%,which were in line with the requirements of biological sample quantification analysis. Except for there was no statistical significance in the absorption of kaempferol absorption in duodenum of model group at different concentrations,absorption of other five constituents in duodenum of normal and model rats increased with the increase of the concentration of active constituents ,and absorption of medium- and/or high- concentration active constituents (except quercetin )in model group was significantly lower than normal group (P<0.05). In normal group ,the absorption of kaempferol was more in jejunum,ileum and colon ,isokaempferol was more in ileum ,vitexin and protocatechuic acid were more in jejunum and ileum , kaempferin-3-O- β-D-glucoside was more in duodenum ,jejunum and colon ,quercetin was more in colon ;in the model group ,the absorption of Polygonum orientale in jejunum and colon was more ,the absorption of isokaempferol in 4 intestinal segments was little different ,vitexin was mainly absorbed in ileum ,protocatechuic acid and kaempferol- 3-O-β-D-glucoside was mainly absorbed in jejunum ,quercetin was mainly absorbed in duodenum and ileum ;in the same intestine ,the absorption of constituents in the model group was less than normal group. After adding verapamil ,absorption of all constituents in the normal group increased ,but the difference was not statistically significant (P>0.05);absorption of kaempferol ,isokaempferol,vitexin,protocatechuic acid and kaempferol- 3-O-β-D-glucoside were all increased significantly in model group (P<0.05),while there was no statistical significance in the increase of quercetin (P>0.05). After the bile flowed into the duodenum ,absorption of protocatechuic acid was increased significantly in normal group (P<0.05);absorption of other active constituents were increased significantly in model group,except for isokaempferol and quercetin (P<0.05). CONCLUSIONS :Six active constituents of P. orientale were absorbed in the whole intestine of normal and MI model rats ,and the absorption of above constituents may be enhanced more significantly by P-gp inhibitor and bile under pathological condition.

METHODS:Five batches of Rhizoma Begoniae from Guizhou were collected ,and the microscopic characteri stics of the Rhizoma Begoniae powder were observed. DOI :10.1002/med.21634. qualitative identification of Rhizoma Begoniae was conducted by TLC ,and the contents of moisture ,total ash and water-soluble extract in Rhizoma Begoniae were determined. The contents of rutin were determined by HPLC. RESULTS :The powder of Rhizoma Begoniae medicinal materials was brown ,stone cells were square ,polygonal-like or irregular. There were many starch grains and few complex grains. The conduit ,calcium oxalate square crystal/cluster crystal were visible. The same fluorescence spots were found in the same location of TLC atlas of Rhizoma Begoniae control herb. The moisture ,total ash ,water-soluble extract contents were 10.15%-11.41%,8.70%-12.59% and 16.91%-19.58%,respectively. The linear range of rutin were 18.47-147.8 μg/mL (r=0.999 8);RSDs of reproducibility ,intermediate precision and stability tests (8 h)were all lower than 3.0%;the average recoveries were 99.39%-100.29%(RSDs were 0.23%-2.59%,n=3);the contents of rutin in 5 batches of Rhizoma Begoniae were 0.102%-0.198%. CONCLUSIONS :The contents of moisture and total ash shall not exceed 13.0% and 14.0% respectively, and the contents of water-soluble extract and rutin shall not be less than 15.0% and 0.080%. The quality standard established in this study can be used for the quality control of Rhizoma Begoniae from Guizhou.

Objective To understand the disease progression and human leukocyte antigen (HLA) gene polymorphism of HIV-infected persons without disease progress for long term,also known as long-term non-progressors (LTNPs),in Henan province.Methods A retrospective study was conducted in 48 LTNPs with complete detection and follow-up information during 2011-2016 in Henan.Changes of CD4 +T cells counts (CD4) and viral load (VL) during follow-up period were discussed.Polymerase chain reaction-sequence-specific oligonucleotide probe (PCR-SSOP) was used for the analyses of HLA-A,HLA-B and HLA-DRB1 alleles between LTNPs and healthy controls.Results From 2011 to 2016,forty-eight LTNPs showed a decrease of the quartile (P25-P75) of CD4 from 601.00 (488.50-708.72)/μl to 494.00 (367.00-672.00)/μl,and the difference was significant (P<0.05).The increase of the quartile (P25-P75) of log10VL from 3.40 (2.87-3.97) to 3.48 (2.60-4.37),but the difference was not significant (P>0.05).HLA polymorphism analysis revealed that HLA-B*13:02 and HLA-B*40:06 were more common in LTNPs (P<0.05),while HLA-B*46:01 and HLA-DRB1*09:01 were more common in healthy controls (P<0.05).Conclusions The CD4 of LTNPs in Henan showed a downward trend year by year.HLA-B*13:02 and B*40:06 might be associated with delayed disease progression for HIV infected persons in Henan.

ABSTRACT

To investigate the value of ventricular tachycardia (VT) score in diagnosing pre-excited tachycardia.
 Methods: Twelve-lead electrocardiograph results were obtained from 30 patients at pre-excited tachycardia attacking stage who were diagnosed by electrophysiology. We scored pre-excitation tachycardia based on the VT score. To analyze the electrocardiogram of pre-excited tachycardia using 7 diagnostic indicators of the VT score and calculate the specificity of 7 diagnostic indicators and right superior axis (-90º to ±180º), the differences were compared among VT score of 2 points and brugada, Wellens, and Vereckei algorithms in diagnosing pre-excited tachycardia. According to the specificity of Vereckei, Wellens, and Brugada algorithms, and VT scores from low to high, their prediction value and differences were analyzed.
 Results: Single indicator such as atrioventricular (AV) dissociation or right superior axis (-90º to ±180º) showed the highest specificity (100%) for identifying pre-excited tachycardia. No patient with VT score was ≥3 points, and the specificity was 100%. The specificity of VT score of 2 point was higher than that of Brugada, Wellens, or Vereckei algorithms in the diagnosing pre-excited tachycardia (76.7% vs 50.0%, 23.3% or 20.0%, P<0.05). The specificity of Vereckei, Wellens, and Brugada algorithms and VT score were gradually increased after each of stepwise individually eliminated VT (20.0%, 40.0%, 66.7%, 83.3%, P<0.05). However, there was no significant difference in the specificity in the remaining false positive cases between the 4 methods and VT score.
 Conclusion: VT score ≥3 points can identify pre-excited tachycardia and VT with 100% specificity. VT score of 2 points cannot completely distinguish pre-excited tachycardia from VT, but specificity of VT score with 2 points is obviously higher than that of Brugada, Wellens, and Vereckei algorithms.

ABSTRACT

<p><b>OBJECTIVE</b>To study a case with weak D59 phenotype identified among ethnic Han Chinese population.</p><p><b>METHODS</b>Routine serological tests were used to analyze the reaction patterns, and the RhD epitopes were verified with 12 monoclonal antibodies. Sequence-specific primer PCR was applied for typing the weak RhD and RhD zygosity in the proband and his family members.</p><p><b>RESULTS</b>A c.1148T>C variant was identified in the proband, for which serological test indicated a weak D phenotype. RHD zygosity testing confirmed that the proband had a RHD+ /RHD- genotype.</p><p><b>CONCLUSION</b>A weak D59 phenotype was firstly identified in a Chinese individual.</p>

ABSTRACT

Objective To investigate the differential MSCT diagnostic features and comparative study of subtypes of renal cell carcinoma (RCC). Methods All of the renal cell carcinomas including 14 chromophobe RCCs (ChRCC), 10 papillary RCCs type 1(PRCC Ⅰ), 15 papillary RCCs type 2 (PRCC Ⅱ), 7 mucinous tubular and spindle cell carcinomas (MTSCCs) were investigated except for clear cell RCC. Dynamic contrast-enhanced CT was conducted in each case after intravenous administration of contrast agent, and the data including all the CT manifestations and the enhancement features were analyzed and contrasted together. Results The indexes including enhancement homogeneity, border of the tumor, renal pelvis violation, blood vessel in tumor showed statistically significant difference between the 4 subtypes (P<0.05), but no difference in the calcification of the tumor. Only the enhancement degree of MTSCC was lower than the kidney medulla in all of the three enhancement scanning phases, while the other 3 tumors' enhancement degree was higher than the kidney medulla in the cortical phase. Peak contrast enhancement of ChRCC was located in the cortical phase, however, peak contrast enhancement of the others did in the nephrographic phase. Conclusions Enhancement characteristics combined CT features is of great help in differential diagnosis of 4 subtypes of RCC.
